Taking NMN does not automatically establish a need for TMG. The sources reviewed here do not establish a universal NMN–TMG requirement or a standard pairing ratio. A methylation explanation is a reason to ask a research question, not proof that every woman needs another supplement. Review the actual ingredients, the human comparison and your clinical context before adding to your routine.
What is the methylation argument?
Using a methyl group in metabolism is not, by itself, evidence of clinically important depletion. TMG means trimethylglycine, also called betaine. Nicotinamide metabolism can involve transfer of a methyl group; that helps explain why people ask whether a NAD+ precursor affects methyl-donor balance. But demonstrating a pathway is different from showing a harmful change, then showing that adding TMG prevents it. Nicotinamide metabolism and the human NR study.
That three-step distinction is the useful starting point: mechanism, observed change, and a tested benefit from the proposed addition. A claim can be plausible at the first step without reaching the third.
What does the human evidence actually test?
An NR trial cannot settle an NMN pairing question, and an ongoing NMN study cannot supply results yet. Here is what each source contributes without transferring findings between products.
| Evidence | What it says | What it cannot establish |
|---|---|---|
| 2019 NR trial | Eight weeks in 140 overweight, otherwise healthy adults. A post hoc analysis found no increase in plasma homocysteine. | That all NMN products are safe, or that TMG is needed or unnecessary for every NMN user. |
| NCT07144527 registration | A recruiting exercise-tolerance study includes NMN-with-TMG, conventional NMN and placebo intervention sequences. No results posted when checked October 13, 2026. | A demonstrated advantage of the combination or a recommendation for routine use. |
| Online combination experience | A person describes what happened while taking a combination. | Which ingredient caused a change, whether it would recur, or whether you should copy it. |
The NR trial was commercially funded and conducted predominantly in white, middle-aged adults; homocysteine was assessed after the main trial rather than as its primary outcome. It is useful context, not long-term NMN evidence. The current NMN study record concerns adults aged 60–80 and exercise-related outcomes. A registration documents a plan and status, not a successful result.
Our NMN versus NR guide explains the ingredient distinction. This article adds the separate question of whether evidence justifies a second ingredient. For Yesnap's broader evidence context, read the Science information.
Check a pairing claim with three questions
Ask for the comparison before choosing the combination. This original claim filter helps separate a biochemical story from a practical reason to add a supplement.
- Same ingredient? Was the research on NMN, NR, nicotinamide or something else? Write the tested preparation beside the product you are considering.
- Same comparison? Did researchers compare NMN alone with NMN plus TMG, or only a combination with placebo? A combination-versus-placebo finding alone cannot isolate TMG's contribution.
- Relevant result? Were results published, in people like you, and for the outcome you care about? A biomarker and a clinical benefit are separate endpoints.

What if a seller recommends matching the amounts?
A simple ratio is not evidence of an individual need. Ask for a human study supporting that exact recommendation and its applicability. The sources above do not supply a standard NMN–TMG ratio. Do not turn a study's dose into a personal serving instruction.
Keep the next step proportionate
You can review NMN without committing to a growing stack. Bring your full supplement and medication list to a clinician or pharmacist, especially when a lab result, new symptom or existing condition prompted the question. Avoid treating symptoms as proof of “methyl depletion” or adding another product to counter an unexplained reaction.
NIH's choline fact sheet describes nutritional needs as influenced by diet and individual factors. That context does not establish a TMG prescription for NMN users. A personalized nutrition or clinical assessment is a different task from comparing two shopping options.

The studies discussed do not establish effects of this finished product or a need to pair it with TMG.
Frequently asked questions
Is TMG the same as NMN?
No. TMG is trimethylglycine, or betaine. NMN is nicotinamide mononucleotide. A shared metabolic discussion does not make them interchangeable.
Does a normal homocysteine result mean I need no TMG?
This article cannot interpret your result or establish a supplement need. The NR study does not answer that individual NMN question.
Is there a proven NMN-to-TMG ratio?
The sources reviewed here do not establish one. Request the human evidence behind a proposed ratio rather than copying it.
Does an ongoing trial prove the combination works?
No. A registered trial shows what researchers intend to test. Check whether results are posted and what comparison they actually support.
